Have to get supplies some time. Boat is generally not the worst idea in the world. But it certainly has its drawbacks. One of the main ones is other people, also on boats, who may be less well supplied. Personally if I had a ship with 12 dudes on it I'd be more intent on raiding other boats than risking going ashore. There is also the point that one containment breach (i.e. one person gets on your boat who's infected) and the whole plan is fucked. Too contained. Similar problem with a lot of people's "high up in tall building" plans.
However, as I say, not the worst idea in the world. Just don't think for a minute it's a full-stop solution.
This is never the case though, there is always a stairwell. But let's take your hypothetical building. You can store what, a month's food at a time? Then you need to go out the fire escape. What if there are 5000 zombies milling about down there? Even a hundred = death.
I don't like fortress survival plans much. I'm on that nomad tip.
